چکیده
Papain-like protease is a SARSCOV-2 that functions for ubiquitination in host cells. Caffeine compounds have been widely reported to antioxidant, anti-inflammatory, antidiabetic, and antiobesity activities. However, hexose malic acid coffee rind as potential inhibitors of the papain-like SARSCOV2 not reported. This study investigated PLPro inhibitor agents inhibiting ubiquitination. In silico studies were used identify two by interacting with (PLPro) proteins Molegro virtual Docker 5 program. Next, ligand-protein complex visualization was done discovery studio version 5.0. showed interactions on several active site residues. The inhibition stimulation interferon compound complexes – protein hydrophobic interactions, hydrogen bonds, van der Waals forces, which contributed formation bond energy strong bonds between proteins. this study, it concluded act protein.

MERS‐CoV papain-like protease (PLpro): expression, purification, and spectroscopic/thermodynamic characterization
Within a decade, MERS-CoV emerged with nearly four times higher case fatality rate than an earlier outbreak of SARS-CoV and spread out in 27 countries in short span of time. As an emerging virus, combating it requires an in-depth understanding of its molecular machinery. Therefore, conformational characterization studies of coronavirus proteins are necessary to advance our knowledge of the matt...
متن کاملA chimeric virus-mouse model system for evaluating the function and inhibition of papain-like proteases of emerging coronaviruses.
To combat emerging coronaviruses, developing safe and efficient platforms to evaluate viral protease activities and the efficacy of protease inhibitors is a high priority. Here, we exploit a biosafety level 2 (BSL-2) chimeric Sindbis virus system to evaluate protease activities and the efficacy of inhibitors directed against the papain-like protease (PLpro) of severe acute respiratory syndrome ...
متن کاملDeubiquitination, a new function of the severe acute respiratory syndrome coronavirus papain-like protease?
A new coronavirus has been identified as the infectious agent of severe acute respiratory syndrome (SARS). Although SARS was successfully contained by quarantine measures, the reconstructed itinerary of the virus through 30 countries and its high mortality rate illustrate the global threat that this newly emerging disease represents (17). During the expression of the SARS coronavirus (SCoV) gen...
متن کاملA noncovalent class of papain-like protease/deubiquitinase inhibitors blocks SARS virus replication.
We report the discovery and optimization of a potent inhibitor against the papain-like protease (PLpro) from the coronavirus that causes severe acute respiratory syndrome (SARS-CoV). This unique protease is not only responsible for processing the viral polyprotein into its functional units but is also capable of cleaving ubiquitin and ISG15 conjugates and plays a significant role in helping SAR...
متن کاملTransmissible Gastroenteritis Virus Papain-Like Protease 1 Antagonizes Production of Interferon-β through Its Deubiquitinase Activity
Coronaviruses (CoVs), such as human coronavirus NL63 (HCoV-NL63), severe acute respiratory syndrome CoV (SARS-CoV), murine hepatitis virus (MHV), porcine epidemic diarrhea virus (PEDV), and Middle East Respiratory Syndrome Coronavirus (MERS-CoV), encode papain-like (PL) proteases that inhibit Sendai virus- (SeV-) induced interferon (IFN-β) production. Recently, the crystal structure of transmis...
